ThruPLEX® DNA-seqKit은 혁신적인 ThruPLEX chemistry를 이용하여 DNA library를제작할 수 있는 제품이다. DNA library는 50 pg 수준의극소량의 DNA로도 가능하며, multiplexing분석을위하여 최대 96개까지의 index를 사용하여 민감도를 올릴수 있다. 본 제품은 샘플 종류에 상관없이 fragmentation된 DNA라면 모두 적용이 가능하다(Genomic, biofluids suchas cell-free DNA, DNA from FFPE materials, cDNA). ThruPLEX® DNA-seq Kit은single tube(well) reaction으로 별도의 정제(purification)과정이나 tube교체가 필요 없으며, 약 2시간이면완료할 수 있다.제품에 따라 12, 48 , 96 single- 혹은 dual-readIllumina-compatible indexes가 포함되어 있으며, index에 따라tube (6, 12) 혹은 pre-dispensed barcodemicroplate에 index가 제공된다. ThruPLEX® DNA-seqKit은 다양한 DNA-seq application과 RNA-seq을위한 cDNA 그리고 ChIP-seq등에사용될 수 있으며, 주요 target enrichmentplatform protocol도 함께 제공되고 있다.

[그림1] ThruPLEX® DNA-seqKit Single- tube WorkflowStartingwith as little as 50 pg of fragmented double-stranded DNA or cDNA, ThruPLEXDNA-seq Kit creates indexed libraries in 3simple steps: end repair, adapter ligation, and high-fidelity libraryamplification. The streamlined workflow prevents sample loss and is performedin about 2 hours in a single tube or well preventing sample loss and enhancingpositive sample identification.
[그림2] ThruPLEX® DNA-seqKit 기술 ThruPLEX® technology is a 3-step reaction that starts with fragmented double-stranded DNA(0.05 ng to 50 ng) which is repaired in a highly efficient process. Backgroundis reduced using double-strandedadaptors with no single-stranded tails. Blunt end ligation occurs withhigh-efficiency. Blocked 5’endsreduce adaptor-adaptor ligation. Background is further reduced by destroyingunused adaptors after ligation.
