□ Tet-On Inducible Expression - The 3rd Generation

The Tet-On 3G Tetracycline Inducible Gene Expression Systems are the 3rd generation of the most powerful, versatile, and widely cited inducible mammalian expression systems available. The 3G system offers a significant improvement over Tet-On and Tet-On Advanced by further reducing basal expression and increasing sensitivity to doxycycline, a tetracycline analogue (Table I, Figures 1-3).

□ Tet-On 3G: Lowest Background Ever, Highest Sensitivity, Highest Fold Induction
Lowest BackgroundMutations within thePTRE3G promoter have reduced background expression from this inducible promoter by 5-20 fold compared to our previous tightest promoter PTight (Figure 1).Highest SensitivityMutations to create the Tet-On 3G transactivator have significantly increased its sensitivity to the inducer doxycycline (Dox), so that at very low Dox concentrations (between 5-10 ng/ml), expression can be 100-150 fold higher when using a Tet-On 3G system, compared to Tet-On Advanced (Figure 3). An overnight exposure to as little as 10 ng/ml of Dox generates very high expression levels (Figure 4).Highest Fold InductionFold induction refers to the difference between the induced and uninduced states. Since Tet-On 3G retains the high maximal expression demonstrated by all Tet-On and Tet-Off systems but has significantly reduced basal expression compared to its predecessors, the fold induction levels are far higher (Figure 2).We have observed unprecedented fold induction levels even in transient cotransfection experiments-up to a 27,000 fold difference between the induced and uninduced expression levels in MCF7 cells (not shown). The PTRE3G promoter results in significantly reduced basal expression. HEK 293 cells were transiently cotranstected with both the response vectors (containing luciferase) and regulator vectors from each of the Tet-On 3G and Tet-On Advanced Inducible Expression Systems. The cells were cultured in the presence and absence of Dox, and after 24 hr, luciferase expression was measured. Although both systems provided strong expression in the presence of Dox, the Tet-On 3G System produces far lower background expression in the absence of Dox (inset).
